Assessment of the successful Transformation of Bacterial Cells with Different Plasmid ABSTRACT Bacterial transformation is one the best strategies available in genetic engineering. The report sought to assess the differences between two plasmid models used in studying bacterial resistance (pUC18 for ampicillin and pBR322 for tetracycline). Bacterial cells were aseptically prepared and transformed by heat shocking, and plated on sets of media with different antibiotics alongside control groups and incubated for colony growth. Results showed that pUC18 for ampicillin reported 172 colonies against 35 colonies counted in the pBR322 for the tetracycline model. These showed that the two models differed in the way competent cells incorporated the plasmids (p Read More

Rapid Colony Transformation of E -Coli with Plasmid DNA

Recovery: LB broth is added to the DNA/cell suspension and incubated at 37C before being put on plates with different selected antibiotic-resistant markers.... The purpose of the paper titled "Rapid Colony transformation of E -Coli with Plasmid DNA" demonstrates that e-coli in the combination with a foreign gene-in this case-plasmid DNA - shows the ability to multiply into an antibiotic-resistant phenotype.... Transformed cells recover from the treatment, amplify the transformed plasmid, and begin to express the antibiotic-resistant strain....
